Where Mastercard differs most usefully from a bank transfer is speed of setup. There is no bank login, no wallet to top up first, and the deposit clears the moment your issuer approves it. Where it differs least usefully is reliability: Canadian issuers decline gambling merchants often, and no amount of retrying changes that.

Mastercard is a payment network. The card in your wallet is issued by a Canadian bank or credit union, and that issuer — not Mastercard, not the casino — decides whether a gambling transaction goes through. This is the single most useful thing to understand about card deposits, because it explains why the same card can work at one site and fail at another.
Debit Mastercard is common in Canada and draws directly from your chequing account. At the cashier it is handled exactly like a credit card, but it sidesteps the cash-advance fee and immediate interest that some credit issuers apply to gambling transactions. If you have both, debit is usually the better choice here.
Prepaid Mastercards are a different matter. They will often fund a deposit, but they cannot receive a withdrawal, so any winnings have to leave by another route. If you use one, set up Interac before you need it.

The pattern mirrors Visa exactly, because these operators treat the two networks as one "cards" method rather than configuring them separately.
Against Visa there is effectively no practical difference at these casinos. Both networks are handled by the same cashier option, carry the same minimums and the same withdrawal restrictions. Use whichever card your issuer is less likely to decline.
Against Interac, Mastercard is faster to set up but less dependable. Interac works in both directions at all eight brands and is not subject to issuer blocking.
